Abstract
Introduction: Ulcerative colitis (UC) is a chronic inflammatory disease caused by an imbalance in the immune system. However, the exact role of IL-10 family cytokines in the development of this disease remains unclear. This study was designed to assess the effects of quinic acid (QA), coumaric acid (CA), ferulic acid (FA), and syringic acid (SA) on IL-10 family cytokines in an experimental model of colitis.
Methods: A total of 128 male Wistar rats were divided into 16 groups (n=8 per group). The groups included control, UC, dexamethasone (2 mg/kg/day), and treatment groups receiving different doses (mg/kg/day) of CA (150, 100, 50), SA (50, 25, 10), FA (60, 40, 20), or QA (100, 60, 30, 10). All phenolic acids were given orally by gavage for 5 days. Expression of IL-10 family cytokines (IL-26, IL-24, IL-20, IL-19, and IL-10) was measured by real-time PCR.
Results: Acetic acid administration significantly decreased IL-10 and IL-19 and increased IL-20, IL-24, and IL-26 mRNA expression in the colon tissue. IL-10 and IL-19 expression was significantly upregulated by dexamethasone, QA (100), CA (150), SA (50), and FA (60) compared with the UC group. IL-20 and IL-24 expression were significantly decreased by dexamethasone, QA (60, 100), CA (100, 150), SA (25, 50), and FA (40, 60). IL-26 expression was also significantly reduced by QA (60, 100), CA (150), SA (50), and FA (60).
Conclusion: High doses of QA, CA, SA, and FA increased IL-10 and IL-19 expression and decreased IL-20, IL-24, and IL-26 expression in rats with UC.
